In Vitro Quantitative Clonal Assay of CFU-Granulocyte, Monocyte in Mouse Bone Marrow

Abstract
|
|
|
To try quantitative and selective culture of Colony Forming Unit- Granulocyte, Monocyte (CFU-G.M) in mouse bone marrow, bone marrows of 56 C57 black mice were extruded frdom both femurs by enforced media infusion method.
Pokeweed mitogen spleen cell conditioned media (PWMSCM) was used as a growth promoting activity in culture. It is easy to make PWMSCM in small laboratory and is considered to be a useful promoting activity instead of mouse lung conditioned
medium.
Agar removal, drying and Wright-Giemsa staining method was used for colony counting and waqs cosidered to be superior to classical methods such as inverted microscopy and in situ staining to count CFU- C. M colonies accurately. The mean number of
CFU-
G. M colonies formed was 36.43/1×106 cells/ dish and I believe this quantitive clonal assay method of CFU-C.M is more advanced method than classical methods and would be helpful for further murine and human in vitro stem cell studies.
